Hi I’m back feeding my house to hook up my generator the outlet box I’m using is metal and has a ground screw would it be ok to tie into the ground coming off my 220 outlet? Or should I run a separate ground wire for this

Both. You’ll need to ground the outlet and the box to the house ground.
